Use "dump database" to backup your database
You can make a backup of the data files but than the database must have no activity (shut down) or use "quiesce database"

Since you are approaching db backup with a SQL server (does it ?) mindset, let me say that Sybase online [without having to take either the users or the server offline] backup facilities and speeds beyond what you are expecting. It will serve you to RTFM (they are available online) and Sybase understand backup and restore options, rather than to try to backup a Sybase db using a primitive (PC or MS) style.

If you do not understand that an online backup of a db is possible, post and I will reply.
